Functional efficiency of cell-engineered liver constructs based on tissue-specific matrix (experimental model of chronic liver failure)

چکیده

Objective: to investigate the functional efficiency of a cell-engineered construct (CEC) liver based on tissuespecific matrix consisting decellularized rat fragments, allogeneic cells and multipotent mesenchymal stromal (MSCs) isolated from bone marrow an experimental model chronic failure (CLF). Materials methods . In creating CECs, for decellularization were obtained from male Wistar rats. MSCs marrow. The efficacy CEC was investigated on CLF by priming rats with CCl 4 solution. At different periods after implantation, outcomes assessed biochemical parameters cytolysis.  Morphological changes in the analyzed histochemical in control (administration saline solution into liver parenchyma) parenchyma) groups. Results It was shown that implantation proposed normalizes blood structural disorders of damaged faster (by day 30 introduction instead 180 control). CEC was also shown have reduced animal mortality 50 0%, which is due early activation proliferation of viable formation new vessels. These effects are down either stimulation of the internal regenerative potential during or long-term functioning transplanted as part matrix. Conclusion. CEC, implanted parenchyma laboratory animals model, has activity.

In situ expansion of engineered human liver tissue in a mouse model of chronic liver disease.

Control of both tissue architecture and scale is a fundamental translational roadblock in tissue engineering. An experimental framework that enables investigation into how architecture and scaling may be coupled is needed. We fabricated a structurally organized engineered tissue unit that expanded in response to regenerative cues after implantation into mice with liver injury. Specifically, we ...

[Acute on chronic liver failure].

Acute-on-Chronic Liver Failure (ACLF) is a recently introduced term defined as severe acute deterioration of an established liver disease. This entity usually develops after an acute insult. The main clinical manifestations are hepatorenal syndrome, hepatic encephalopathy and organ failure, with a high risk of death in the short term. The true incidence of ACLF remains difficult to determine du...

Acute-on-chronic liver failure.

Acute-on-chronic liver failure combines an acute deterioration in liver function in an individual with pre-existing chronic liver disease and hepatic and extrahepatic organ failures, and is associated with substantial short-term mortality. Common precipitants include bacterial and viral infections, alcoholic hepatitis, and surgery, but in more than 40% of patients, no precipitating event is ide...

Acute-on-chronic liver failure

Acute-on-chronic liver failure (ACLF) is an increasingly recognized distinct disease entity encompassing an acute deterioration of liver function in patients with chronic liver disease. Although there are no widely accepted diagnostic criteria for ACLF, the Asia.Pacific Association for the Study of the Liver (APASL) and the American Association for the Study of Liver Disease and the European As...
